I have a MAc Pro 13", a iPhone 4S and a Mac 27". Do I have to buy a copy for all three? to play on all three.

Software (and games) purchased on the Mac App store can be downloaded on multiple macintoshes - you do have to use the same iTunes account on each machine, of course.
The iPhone is a different platform entirely - no Mac software will run on it and vice-versa. Also note that the iPhone version of Ticket to Ride is focused on solo game play - and will not let you play with the copies you may purchase for your Mac.
